Abstract

Objective: To review withanolides as bioactive constituents that may influence stress-response signaling pathways. This mechanistic paper summarized in vitro, ex vivo, and animal findings related to HPA-axis modulation, inflammatory signaling, oxidative stress, and neurotransmitter-associated pathways. Withanolide-rich preparations were reported to alter stress-related biomarkers and to interact with cellular pathways relevant to resilience, including glucocorticoid signaling and inflammatory mediators. The review highlighted that many observed effects may arise from a combination of immunomodulatory and neuroendocrine mechanisms rather than a single target target. Because the evidence base is largely preclinical and extract compositions vary considerably, direct comparisons across studies are difficult. Nonetheless, the consistency of effects across stress models makes the pathway-level hypothesis plausible. The paper positions withanolides as candidate adaptogenic compounds and underscores the need for standardized chemistry, dose reporting, and human biomarker studies before stronger efficacy claims can be made.
